No, once a post is locked, you can't post to it anymore... what you witnessed was that the post lock status hadn't been updated across all servers yet. There is a small window after a lock where it technically is possible to post to a thread, but it is very small, and afterwards, even URL manipulation won't work.

This one is pretty insignificant:
1. I clicked "reply" to reply to a thread.
2. As I was authoring the reply, a mod locked the thread.
3. I clicked "submit" and my post was added to the thread after it had been locked.
So locking the thread doesn't literally disable posting, as much as it just eliminates the presence of "reply" buttons. That might be fine with you, but I just thought I'd mention it in case you were interested. This is the thread.
Edit: Actually I just attempted to post to a locked thread (For test purposes) by URL manipulation, and received the "This thread is locked, you don't have permission, blah, blah" screen, so I don't how I was able to post in the above thread. *shrug*
