As stated in Halo: The Fall of Reach, another plan for the entire group of Spartans was planned and they had just started on their way when a distress signal came out from Reach. The squad was split up into two groups. MC, Linda, and James went off on what they considered the most dangerous of the two missions because it was in a zero-gravity environment, whereas the rest were sent to Reach because Spartans ALWAYS won on the ground. You may or may not see Linda or James. I hope this encounter is part of the beginning of the game.
Master Chief believed himself to be the only Spartan to live after the incidents of First Strike, but this is not the case.
The remaining Spartans were dropped to the surface on a hot burn, ODST style. Helljumpers do not always make it. This would also make another awesome beginning to a level in this game. This would also bring up another question in my mind: would we be able to hone in on the specific Spartan's strengths and/or weaknesses? Will we play as ODSTs for part of the game?
The fight will be amazing, if anything like depicted in the book. Perhaps we will see the face of Dr. Halsey? The beginning of this fight will hopefully be amazing and action packed. The jobs the Spartans were given should make for interesting fights.
I don't have the time to check dates. It is possible to find the dates of these battles and compare them to dates listed on helmets in the information listed in Halo 3 to determine if certain helmets could possibly be used. If so: Would the helmet change a certain aspect of gameplay depending on the Spartan you could possibly be playing as? Certain Spartans preferred particular gear. Linda was a sniping expert and customized her rifle. By my own guess, judging by the header picture in this forum, each Spartan character will have unique equipment, with different subtleties.
The specifics of who is in Gray team are given in the book The Cole Protocol. As far as the timeline is concerned, it took place before the events at Reach. They consisted of Adriana-111, Jai-106, and Mike. They operated on covert missions alone, therefore are probably left out of this game.
Another team mentioned, Green team, could also be part of this game.
As far as the implications of Spartan III's in this game, that would be impossible. They were implemented and used after the fall of Reach. They were kept a secret. This also means Kurt will not be involved. Read The Ghosts of Onyx for more details.
Since MC always led Blue team, we are most likely left with the other option we've been shown in the book: Red team. Which segment(s) of Red we will see is unknown. It is also possible MC will make an appearance and be playable in Reach.
There are alot of questions that we won't be able to answer or even make educated guesses on until Bungie clues us in with more hints. They've been known to be very subtle, so I'm sure fans like myself will be scocuring any media put out for anything.
