WHAT TO EXPECT IN REACH
Bungie has given us reason to believe the game will be squad-based. There is little evidence to support this, but is reasonable to believe some vehicles from Halo Wars will make an appearence, such as the Cobra, and the Hawk.
Some Covenant vehicles might be thrown into the mix, too.
GAMEPLAY
There is evidence to suggest that Reach's gameplay will be class-based, like CoD. More details here.
ABOUT THIS THREAD
I will be using the picture from the top of the page to give you visual detail. (The Forum Picture) Please keep in mind that these are ESTIMATED GUESSES, and are not one hundred percent accurate, although I do have evidence to back them up.
NEW SPARTAN'S
In the Halo: Reach Teaser Trailer, a few Spartan's are introduced. Throughout the books, especially the Ghosts of Onyx, there are multiple attempts at more SPARTAN Projects, to breed more Spartans. However, the known Projects take place after the events at Reach. In the trailer, a Spartan speaks up, claiming hiself/herself as Sierra 259. That is confusing, seeing as the original Spartan's number from 1 to 150.
Of course, Sierra may not be referring to a Spartan, but it is still likely.
You decide?
Therefore it is suggesed that this could be possible evidence of a hidden SPARTAN Project not discussed in the books, as well as suggesting new playable Spartan's.
CHARACTERS
(See the picture above)
NOTE! I could be WAY off on these Spartans. Although I have evidence, I do not have proof.
In the middle is none other than the Master Chief himself, or John 117. In the book, The Fall of Reach he is described as the Spartan team leader, so it's only logical he'd be in the center.
To John's right would have to be Fredric 104. He is recognized for his skills with a combat knife, which the Spartan in the picture is obviously holding.
To Fred's right is Linda 058, armed with an SRS Sniper Rifle. In the books, she's known for her deadly aim with Sniper Rifle's, so it's only logical it's her.
To John's left is Kelly 087. The picture gives evidence of a slender figure, supporting the body of a woman. Kelly is known for being shorter than most Spartans, and known for her incredible speed.
To the far left is Samuel 034, who is described as the tallest Spartan. (Notice he is much, much taller than any of the others in the picture) He dies before the events of the Fall of Reach, so it is suggested that the game's plot will take place before Reach, as well as on it.
Cortana was introduced in The Fall of Reach, right before the Spartan's got their new armor. (The ones with sheilds) She might be in the game a bit, but it isn't likely.
Gunnery Sergeant Edward Buck was said to be on Reach during it's fall. Also, Bungie has offered Nathan Fillion (Buck's voice actor) to tackle a role in Reach, but he hasn't decided. Dare could be linked into the mix, too! YAY!
Captain Keyes might be in Reach, too. The events take place right before Halo: CE, so he also might be in the game, seeing as the Master Chief escapes aboard the Pillar of Autumn.
And finally, Sergeant Johnson. He might be in the game, believe it or not! He's at the very end of The Fall of Reach, so he may make an appearence! Keep those fingers crossed!
STORYLINE
As mentioned in the Characters section, Sam is dead, and dies before the events of Reach.
Therefore, it is logical to conclude that there may be a short introduction towards the beginning of the game where you train a Spartan, or watch a cutscene about it.
This also suggests that you may fight against the Insurrectionists, a branch of terrorists that the Spartans fight in the books, as well as the Covenant.
There may also be a mission where Kelly, John, and Samuel enter a Covenant Carrier, and destroy it. (The Carrier where Samuel dies)
As they entered and explored the frigate, Sam was struck in the side by an overcharged bolt from a Plasma Pistol wielded by a Kig-yar soldier while trying to protect John from harm. The shot burned a sizable hole in Sam's unshielded armor. The Spartans managed to make it to the engine room of the ship and planted several ANVIL-II ASM missiles on the fusion generator; however, their only means of escape was an EVA, but this was impossible for Sam since the vacuum seal in his armor was compromised. Rather than die in the vacuum of space, Sam volunteered to hold off the Covenant forces and allow the missiles to explode, destroying the entire ship. John later marked Sam as MIA because "Spartans never die"; it is believed that the morale of the public and servicemen alike would fall dramatically if they found out the legendary super-soldiers could actually be killed. - Halopedia
There is evidence that the game will just highlight the events of The Fall of Reach completely, but it is not certain.
Also, I have come across suggestions that the planet at the end of Halo 3's Legandary Ending cutscene is Reach. That is highly unlikely, due to the fact that Reach was glassed by the Covenant. If it was Reach, there would be no lights on the surface of the planet, seeing as all Human life had been extinguished. It is said, however, that instead of Reach, it is Onyx.
However, it cannot possibly be Onyx, since the planet was destroyed. More evidence here.

I hope these have answered any questions regarding Reach. I will try to update this as I come across any more information.
