No one knows what to expect from Bungie's new game, Halo: Reach. All we can do is speculate until the Beta sneaks up and is made public. When exactly will be in the Beta? Nobody knows.
Here are a few core speculation thoughts surrounding Halo: Reach.

PLEASE NOTE: This is purely speculation and nothing else.

STORYLINE
Halo: Reach takes place a few moments after the invasion of Reach. The Circumference contains unsecure nav data on all of Earth's colonies, including Earth herself. By watching the Halo: Reach trailer you can tell that Master Chief and his team are attempting to destroy the nav data, which means that the rest of the Spartans are on the surface. Bungie announced a while back that we won't be playing as any of Master Chief's Spartans. So therefor, the Spartans we'll be playing in the game aren't Spartan-IIs. It is possible that we may be playing as Spartan-IIIs.
In the novel Ghosts of Onyx, Colonel Ackerson founded the Spartan-III project (in protest of Dr. Halsey most likely) to be fielded as cannon fodder to deal with suicidal missions. Since Reach is on the verge of collapse, the only reason these new Spartans in the game would have a top-secret mission to go onto the surface of Reach is if it was a suicidal mission ( a suicidal mission appropriate for Spartan-IIIs). This may also very mean the Halo: Reach will tie into the story of Ghosts of Onyx. The top-secret mission mentioned earlier may be to rescue Dr. Halsey and the remaining Spartan-II soldiers. The rest should be obvious.

EDIT: Tucker 051 informed me that Bungie never said that we wouldn't be playing as SPARTAN-IIs. My fault on my part. I'll make changes to this post as soon as I have gathered more and appropriate material.

TECHNOLOGY
Since Halo: Reach takes place during the time that planet Reach is still the UNSC's main naval yard and hub for scientific activity, it is most likely that we'll see the UNSC at the top of their game and be fielding their best vehicles and weapons (standard and experimental) to fight the Covenant.

Let's take a look at the technology that might be used by the UNSC Marines in Halo: Reach. The Marines stationed on Reach are the war's most veteran soldiers with the latest cutting-edge technology at their disposal. The UNSC Marines will probably have the original armor we've seen in Halo: Combat Evolved, seeing how the Marines aboard the Pillar of Autumn were a detachment of Marines from Reach. The Marine's arsenal may also include vehicles and weapons from Halo Wars since Halo Wars represented humanity at the top of their game.

Now for the Spartans. We'll probably be seeing a lot of cool weapons, vehicles, and armor permutations from the novel Halo: The Fall of Reach in Bungie's new game, as well as some stuff that we've never seen or read about before. This may include the UNSC's own version of the Plasma Rifle that was still in the experimental phase and never "officially" field tested.
Who knows? We might also see the older versions of the Mark-V armor and possibly some Exoskeleton Battle Armor as well. This may include Spartan-III armor.

MULTIPLAYER
There is no doubt that an updated and innovated version of the Halo 3 matchmaking system will be used and a new Firefight mode will be created. However, any new modes is unknown and therefor difficult to speculate. Only when the Beta is released will we truly get a taste of this new online experience.

CONCLUSION
Once again this is purely speculation based on what has been released to the public. Overall, I can say that Bungie will now doubt create a game that will make their classic, Halo: Combat Evolved, wet itself. If even a small portion of this speculation is true, then Halo: Reach will no doubt surpass all other Halo games in every aspect.

From watching the trailer how do you know that the Cheif was destroying the NAV data? There is no evidence to support that.
Bungie also never said that we wouldn't be playing as Spartan IIs, they just said that we wouldn't be playing as MC.
Spartan IIIs were never on Reach.

As I said, this is purely speculation. Please don't take this too seriously. Also, by watching closely, you can spot a Spartan drifting away from the station in the trailer. This is speculated to be the SPARTAN-II named James. Therefor, it is the station that contains the Circumference and the nav data that Master Chief and his team need to destroy.

did they really comfirm that we won't be playing as masterchief? if so im very very very sad.........
