The more spires you destroy during a campaign the more glyphs will appear.
Not real. It would be kinda stupid to ask you prove it, but let me just say that there is no found/proved connection between campaign progress and Glyphs. The reason it could seem like this is because the games random loading nature; it loads many things in random ways.
There are multiple types of glyph and they seem to appear around the campaign waypoints in Mombasa streets and inside the superintendent posts.
Well, that one was right. They are also found near Dead elites and sometimes near audio logs.
All I have managed to really workout at the moment is that they are directly related to the mission associated to the waypoint near the glyphs.

There are also areas through security doors that contain glyphs and engineers, so far I am unable to fathom the relationship of these Glyphs to the other glyphs despite them being the same.
This would be a good moment to list the glyphs.
The most commons Glyphs; let us call them A and B glyphs. As said, they are the most common ones, seen near Waypoints and inside supply caches. There only these two "normal" glyphs; others similar looking ones are the rotated versions of these.
Simple glyphs: The ones seen near Dead Elites. ontain only the Glowing Y-shape, without any other symbols.
These are the...normal glyphs, shal we say. The following ones are more rare, some that only apper in one certain place.
A variation of B glyphs is SI[SuperIntendent] B Glyph. Same as The B glyph, expect that one of the symbols is replaced with an SI face. Found in the Plaza similar to Tayari Plaza and near some Audio Logs, if I remeber correctly.
Sad SI Glyph: One of the two unique Glyphs. Has no other symbols inside it but a Sad SI face. Is only seen in Squidhenge, a place accessed via security doors, has some nice stone slabs and Engineers...But that's another story.
And the most interesting one of the Glyphs: The SI Tri-Glyph. It has three SI faces inside it. Unique. Only seen in the Plaza, where the SI B Glyphs are. Loads randomly; At times, one can't see it at all, but after he has runned around a little bit, it may show up. This happens because of the Random nature of the game. The randomness also causes the Glyph sometimes to change into a SI Di Glyph or SI Mono glyph. Monos are also seen in the Legendary Ending.

I beleive the glyphs may contain some sort of information that could be useful.

More important than the glyphs are the audiologs. I have 27 at the moment and it appears that they the audio logs may conatin the key to understanding the plot line properely and may contain something about the glyphs.
It has been under investigation...No connection is found, however. Keep on it!
Finally the more engineers you kill, the fewer glyphs you get.
