it's highly unlikely that earth was the least human world because there were hundreds and the Cole Protocol aplied to all colonies

the war lasted 30+ years. The covenant eventually glassed Reach, effectively cutting of any other colonies exsisting out there from Military support. If the Covenant didn't get them, they were on their own, and unless a UNSC ship came, they couldn't communicate with other cvoloies unless they went into spaceships themselves, and I doubt that civilians were brave enough to do that.

So there could be others out there, but not able to communicate.

They could communicate without actually flying there. They had near-light speed communication.

On Topic: My guess is that they didn't literally glass every other colony, just all of the significant ones like Reach and Harvest.

The UNSC had over 800 colonies, and only 76 were glassed. ( Play Halo 2, High charity to count the glass. )

The UNSC in terms of size was no where near defeated, but they would of lost their command and many ships if Earth fell.

Where in High Charity is there glass, and where does it say that this glass has anything to do with the number of glassed planets?

The room in First Strike is completely different and is nowhere to be seen Halo 2.

Earth wasn't the last Human world but it was the most important one, what with it having UNSC headquarters on it..

It wasn't the last even if it made sense. There's bunch of active inner colonies, Sigma Octanus IV survived, and it is suggested the Covenant had to skip most of the outer colonies in order to find Reach and Earth.
