Cortana.
No, I'm not joking. In Halo 3, she was already in the first stages of rampancy. Usually, most smart AI begin to go rampant around their seventh year, and Cortana is still rather young for an AI, but for an artificial intelligence, the amount of data they transfer reflects their age/how quickly they become rampant. And, after collecting so much information from the first two Halo installations, and being corrupted by the Gravemind, I think it's safe to say Cortana's time is coming to and end. Plus, what better way to end the new trilogy than to destroy the one character you've grown most attached to in Halo 1, 2 and 3?
Of course, Cortana, while she could be the lead antagonist, won't be the only enemy. I doubt we'll see Forerunners themselves, but I can imagine fighting Sentinels--and I honestly believe the Flood and the Precursors will be making an appearance again. Only time will tell. The way I see it, Cortana will be an ally all throughout Halo 4, halfway through Halo 5 she'll begin slipping, and in Halo 6 she'll be the antagonist we have to destroy.
I can imagine her getting into the shield world's (atleast, that's what it appears to be) systems and pitting an army against the Chief, and I can also see her becoming the new Gravemind, assuming the Flood are back. So, in the end, I can see...
UNSC + Covenant (Chief and Arbiter?) VS Precursors VS Flood and Sentinels (Cortana)
Ah. I also believe the UNSC will be making a reappearance due to the Chief's new armor. (not the armor in the trailer, but the totally redesigned armor in the Halo 4 wallpaper and box art) It had to have come from somewhere. And where there's UNSC, there's Covenant.
Again, this is all speculation. I don't have any hard evidence to back this up, but this is my belief.
