No, you don't have to save them. It makes no difference in the game. It is just more of a challenge or something.

Nope, you don't. Just an added bonus.

If all the marines die, cortana says something, I think.

actually, it does affect the game

it doesnt affect that level, BUT, on the next level where you go and get Keyes from the Truth and Reconcilliation, you get less reinforcements if you dont save each set of Marines at each point

i when i did it on Legendary by myself, not a single marine survived and on the next level, apart from the Marines that you go in with, i got no reinforcements, even when the first lot were wiped out

You NEVER get reinforcements on Legendary once in the ship. Never.

Actually, when you break the Marines and Keyes out of jail they become reinforcements...stupid, retarded, no good reinforcements, but reinforcements nonetheless.
Oh, yeah, plus the guys who take the Grav Lift up with you.

The Rip Saw, i never said IN THE SHIP, i said on the level

and yeah, i forgot about the guys who come with you INTO the ship

but, if you lose all your guys BEFORE you secure the grav lift, if you saved marines previously, a load will be dropped off

if you didnt, they wont
