IIIs are expendable. Basically go on suicide missions.

IIs are better. Quantity over quality. 300 Spartan IIIs were just completely wasted in the beginning of Ghosts of Onyx. IIs are much more helpful.

I wouldn't really call it "better" training, but rather harsher or more extreme. The S-IIIs had 2 or so less years of training, but to make up the difference they trained them a little more intensely. The S-IIs had 2 years more of training, but it wasn't packed or rushed quite as much as it was for the S-IIIs. I would say the overall effect of the training was the same.

S-III's were also all volunteers. They were all orphans of glassed planets and they were asked if they wanted revenge on the covenant.

The III's were also not as large as the II's. Mind you this can be attributed to the fact that no spartan III, save for Noble Team, has made it past the age of 20.

The III's are also more unstable from a personality standpoint. The compound that Kurt came up with made the III's tolerance for pain extremely high. But it also made them more emotionally unstable and aggressive. This isn't that big of an issue as their missions require excessive aggression, but they can lose a handle on stressful situations.

As far as combat effectiveness, aside from their armor, they're pretty comparable to II's. but in my opinion, the II's experience and armor set them apart from the III's.

The biggest difference?

The price tag.

And this is the important one. While Spartans were immensely valuable they coated more than a battle fleet. Quite a hefty price tag for something that could be killed by a few lucky plasma shots. The IIIs were designed to have the pros of the IIs without the massive price tag. That is the main reason they got SPI instead of MJOLNIR.

Spartan 1's were originally ODST's that went through specialized training and piloted a mech-type exosuit that was tied in to a direct power source (usually a generator) S-1's could bring more firepower to bear as the load handling abilities of the exosuits was quite large - however the need for a mobile derivative caused the Spartan II program to be initiated.

From Halo Nation: "In 2491 the Colonial Military Administration relaunches ORION in secrecy, the new goal of ORION is to learn from the mistakes of the previous attempt, and create a soldier unlike anything before it - and to deploy these soldiers behind enemy lines and quell insurgences in their infancy, or destroy established insurgences by destroying their chain of command and expand outward. The Project is headquartered in one of the many orbital docks on Reach, the candidates are, once again, selected volunteers from all Special Force branches of the military. -Better Armor
-More experienced.
-Better genetics.

Spartan III's
-Numerical advantage.
