I would check to see what your specs are, particularly on your GPU. If you have an integrated chipset, and especially if its higher end, it's sharing the RAM with the computer. Most chipsets will take up about 120MB of RAM to itself, meaning that for me, for example, I only 380MB of my physical 512MB avalible. If this is the case, then all you can do is buy more RAM.
Second, do a few virus/adware/spyware scans on your computer. That should eradicate any memory-stealers from your system.
Finally, if to no avail, Ctrl-Alt-Del, go to Processes, and find out what the heck's clogging it up. Pay attention to the right hand column and see what's taking up all the RAM. System standby doesn't count, btw.
